I have lighttpd installed on my workstation (photo organizer,...) and bound to localhost.
I'd like to know if it is possible to access the web server from MacOSX via mol and some configuration hints if possible.

i dont know if it works when bound to localhost. in theory mol communicates with your host over a tun device. that means that gentoo gets another network device tun0 (probably) with an ip address assigned. for me that was 192.168.41.1. mol is using the mol network driver to be the other tunnel endpoint using an ip on the same network (eg. 192.168.41.2). the network communication is done through these 2 endpoints so i guess you probably need to bind your httpd to the tun0 device too to access the http.
